Our goal was to study the effect of mTOR signaling pathway on global levels of Histone acetylation in HeLa cells. We knocked down mTOR in HeLa cells by siRNA and used phosphorylation of S6 protein to examine the activity of mTOR in cell lysates. We probed S6 protein levels by using S6 antibody in western blot which served as a loading control for pS6 levels.

A clear specific band was observed at 32KD corresponding to S6 ribosomal protein in scramble siRNA treated HeLa cells and mTOR siRNA treated HeLa whole cell lysates.Overall, an excellent antibody for western blot which gives a single specific band without any non-specific bands in HeLa whole cell lysates
