PDL-1 is an immune checkpoint molecule that is upregulated in tumor cells and suppress immune detection by interacting with the PD1 receptor expressed on T-cells. Given this role, numerous checkpoint inhibitor molecules/antibodies have been developed, and have shown promising results as anti-tumoral therapeutics in a subset of cancers. Nonetheless, some patients that receive these checkpoint inhibitor molecules develop resistance and require alternative approaches. Therefore, identifying alternative cellular targets that regulate PDL-1 expression on tumor cells are important as they may provide an alternative or additive therapeutic option to conventional PDL-1/PD-1 therapies.Here, we utilize the colon cancer cell line HCT116 to examine PDL-1 and track the expression of PD-L1 using immunoblots. We stimulate cells with the IFN-gamma cytokine to induce PDL-1, which is a way to mimic T-cell induce PDL-1 upregulation in the tumor microenvironment.

Three different samples were run on a gel in the absence or presence of IFNg (100ng/ml -24hrs). As shown, IFNg induces PDL-1 expression as expected in all three samples. Detects the basal and induce forms of PD-L1 well.
