This kit is useful for producing accurate and reproducible results. We have used this kit for gene expression analysis and viral copy number analysis. Our lab has used this kit since 2012 and has not deviated since. This kit is to be used under fast thermal cycling conditions.

Protocol Overview

This kit is optimized for use in SYBR green based RT-PCR. Before beginning, area must be free from contaminants. To set up a reaction, simply prepare a PCR mastermix containing provided reagents: 2x SensiFAST, 10uM forward primer, 10uM reverse primer, and pure water. Pipette into wells, add template, and let the reaction run on a fast thermal cycler.Note: kit comes premixed with ROX (5-carboxy-X-rhodamine, succinymidyl ester) for machines that can read ROX fluorescence.

Tips

Limit talking while preparing the template! Beware of anything that could potentially fall into your plate. Do not use around amplified DNA. It is recommended to use primers with a melting temperature of 60 degrees with optimal amplicon length at 80-200bp.

Results Summary

CT values are exported and used to calculate gene expression levels and or copies of virus. Results are reproducible through different runs.
