My goal was to compare the protein expression of IFN-y in the brains of wt and our newly generated mutant mouse strain. We selected this product because we had great results with the IL-6 kit from this company.

I followed the manufacturers protocol with one exception. I incubated the samples overnight at 4 degrees Celsius rather than the suggested 2 hours at room temperature and also blocked overnight. I added the capture antibody overnight, blocked with 1% BSA in PBS overnight, then incubated with the samples and stds overnight. After adding the Strep-HRP and peroxide substrate solution, the reaction was stopped with 0.86N H2SO4 after 10 mins. and read at 450 nm
Blocking overnight producted the least background
A clean Std curve was generated and IFN-y expression significantly increased in the brains of our mutant mice
