Knockdown of MYC using shRNA plasmids
shRNA plasmids designed against MYC and 293T cells
I used the MX300p to access the level of knockdown of MYC using shRNA constructs.
The most important thing to take care of is to select the right dye present in the mastermix.
48h post-transfection, I prepared the RNA and then cDNA from cells transfected with scrambled shRNA and the shRNA against MYC. The cDNA was used to perform RT-PCR to check the efficiency of shRNA constructs in knocking down the MYC protein.
The machine is fast and the assay is completed in approx 1h10 mins (if the melt curve is not used)
