Cell viability dye is commonly used together with other fluorescent labeled antibody in flow cytometry analysis to discriminate dead cells from live cells. In the current assay, we tested the quality of the Ghost Dye Violet 510 (Tonbo biosciences) using mouse lymph node cells. We found that addition of this Ghost Dye Violet 510 at 1:300 dilution clearly distinguish dead cells from viable cells.

Protocol Overview

Single cell suspension of mouse lymph nodes was prepared and preincubated with Fc block for 20 min in FACS buffer (PBS/2mM EDTA/2%FCS). The cells were then stained with the Ghost Dye Violet 510 (Tonbo biosciences, 1:300 dilution) and PE-CD4 for 30 min in FACS buffer. After washing with FACS buffer, cells were acquired at BD LSRII.

Tips

Warm up the Ghost Dye Violet 510 (Tonbo Biosciences) to room temperature prior to each use.

Results Summary

Addition of the Ghost Dye Violet 510 during staining with fluorescent antibodies can clearly discriminate dead cells from viable cells, as shown in both PE-CD4+ and PE-CD4- compartment.
