The general aim is to explore some rare cells in bone marrow that are CD45-. CD45 depletion was performed to remove most cells unwanted. The reason this product was selected was because it was half the price compared with competitors and of good quality.
Depletion of CD45+ cells from whole mouse bone marrow
Freshly isolated mouse bone marrow
Followed the protocol of Biolegend.
You can perform twice with half the amount recommended each time and get a better result.
Up to 95% CD45+ cells were removed. I have repeated several times and the results are constant. As you can tell from the figure, CD45- cells % increased from 3.36% to 55.9%, namely, 15 out of 16 CD45+ cells were removed, ~95%.

You need a strong magnet that can perform bulk depletion.
