MMP-9 is a matrix metalloprotease produced from immune cells including macrophages and neutrophils, which plays important roles in wound healing, immunity, and angiogenesis. In this product review, I report results that the anti-mouse MMP-9 antibody from Cell signaling showed unsurpassed performance in detecting the protein in both Western blotting and immunofluroscence microscopy.

Results Summary

In the attached figure A, neutrophil total lysate (lane 3) and the supernatants from neutrophils stimulated with low (lane 1) and high (lane 2) amount of PMA were analyzed using Western blot analysis. Mouse MMP-9 protein was clearly detected with the expected size (between 90 and 100kDa). In the figure B, this antibody also successfully detected MMP-9 proteins in PFA-fixed neutrophils using immunofluorescence microscopy.

The Bottom Line

Cell signaling produces a lot of reliable antibodies, which are frequently cited in research articles. This antibody is one of them. I found this antibody worked well both in Western blotting and staining of fixed cells. One more good thing about antibodies from Cell signaling is that they are provided in a glycerol-containing buffer so that they are kept unfrozen at - 20C, which significantly lengthens their shelf-life.
