Subcellular fractionation was performed to separate cytoplasmic and nuclear fraction. GAPDH was used as a loading control for cytoplasmic fraction. GAPDH was present in the first 5 lanes (whole cell lysates), and last 5 lanes (cytoplasmic fractions), but not in the middle lanes (nuclear fraction).
