Leica confocal microscope
Frozen section of mouse spinal cord was rinsed, permeabilized with 1% Triton for 1 hour, blocked in 10% normal donkey serum and stained with goat anti-ChAT antibody (MilliporeSigma AB144P dilution 1/100) for 2 hours. Secondary donkey-anti-mouse Cy3 conjugated antibody was applied for 1 hour. Results shows positive staining in ventral horn motor neurons (with a lot of primary afferent terminations). This is an excellent antibody.
