The goal of this experiment was to detect rate of protein synthesis in C57 mouse bone marrow cells. This kit was selected as it allows labeling of OP-Puro using an AF647 tag.

Mouse bone marrow cells
C57 mice were injected with 100uL of a 50mM OP-Puro solution and rested for 1 hour. PBS injected mice were used as controls. After 1 hr, bone marrow cells were harvested, fixed with 2% paraformaldehyde and permeabilized with saponin solution. OP-Puro was fluorescently labeled using this kit according to manufacturer's instructions.
Kit manual suggests using a 500uL volume to stain, however, this can be reduced to as little as 100uL.
Using this kit, OP-Puro was successfully detected in the APC channel in mouse bone marrow cells.
