Our lab utilizes this system to detect HIV infection levels using virus that expresses luciferase. The assay system is very sensitive and easy to get up.
HIV infection detection
Uninfected human peripheral blood mononuclear cells
Spinfect PBMCs with HIV, allow infection to proceed over a period of several days, dilute lysis solution in water and add to cells, place cells at -80*C for 2 hours, shake lysates at room temperature for 30 minutes, add 20ul lysate to tubes, prepare 1x luciferase assay reagent
Add more of the lysis solution to cells in case samples need to be re-run
This assay system provides highly sensitive readouts for luciferase and can be used in a variety of applications.
