Flow cytometry is a staple method used in our lab to analyze immune cells. We routinely use multiple colors to examine various markers, cytokines, and/or transcription factors in our samples. The compensation beads provide a quick and reliable way to perform compensation without having to use actual cells/samples.

Stain cells with flow cytometry antibodies for surface markers, cytokines, transcription factors, etc., as you normally would. Before analyzing cells, set up compensation controls by adding 100-200ul FACS buffer to flow cytometry tubes, add one drop "Negative Control" + one drop "Anti-Mouse Ig, k" to each tube, add 1-2uL of antibody to tubes (one antibody per tube) to make compensation controls for each color you are using.
