Our lab needed a reliable DNA extraction method to extract >100mg tissue with high yield and high purity, an amount that exceeds the capacity of most commercially available DNA extraction kits. This kit allows for extraction of larger size tissue samples and gives great yields with high purity.

Protocol Overview

Harvest tissue, flash freeze, mince with razor blade, add cell lysis solution and proteinase K, incubate at 65*C for 1 hour then 56*C overnight, proceed with RNase A digestion, protein precipitation, isopropanol precipitation, ethanol wash, and DNA resuspension

Tips

Scale the volumes up or down as needed depending on the size of tissue being extracted

Results Summary

High DNA yields (>500ng/ul in 400uL) and 260/280 ratios at or very near 1.8, 260/230 <2.0
