TATA binding protein is a ubiquitously expressed nuclear protein that binding to the TATA box, upstream of the promoter. It is a part of the transcription machinery of the cell. In our experiment, we detected its endogenous levels in three different cell lines: Hela, MCF7, and SKBR3 by Western. We used it as an internal control for nuclear proteins.

Various nuclear lysates of different cell lines
1:1000 dilution prepared in blocking solution, incubated overnight at 4 degrees
5% skimmed non-fat milk in PBST20
1:5000 anti-rabbit HRP antibody, incubated for an hour at RT

We observed a single band of approximately 40 KDa in all three cell lines, without any noise.
