Dendritic cells (DCs) present antigen to initiate a variety of T cell responses. Distinct subsets of DCs specialize in the induction of immune responses to distinct subtypes of pathogens. Two key subsets have been identified in mice, cDC1 and cDC2, and we are interested in the transcription factors that regulate their development. The CD11b antibody was selected for use in a multi-color flow cytometry for the detection of several DC subsets under different conditions.

Bone marrow progenitors were obtained from the tibia and fibula of healthy mice. DC differentiation was initiated by culture with Flt3L. At day 9 of culture when mature DC subsets have developed, cultures were stained to identify and characterize DC subsets in the contexts of different drug treatments. Amongst CD11c+ DCs, a clear CD11b+ positive population was detectable, representing the cDC2 subset. Signal was retained after aldehyde fixation.
