Stimulation of T cells induces activation and proliferation. Successful stimulation requires signaling through co-stimulatory receptors, including CD27/CD28. We are interested in the expression of costimulatory receptors by human T cells subsets in the blood and how this correlates with transcription factor expression. In particular, heightened expression of CD27 amongst memory CD8+ T cells is associated with a more central memory phenotype. We selected this CD27 APC clone for use in a multi-colored panel to analyze steady state T cell populations.

Results Summary

Healthy human PBMCs were stained for analysis of naive/memory populations within the CD4+ and CD8+ T cell compartment. We focused on CD8+ T cells and assessed CD27 / CD45RA receptor expression. A CD27+ population could be detected when viewed alongside CD45RA expression. However, separation was usually a smear and required the second marker to truly differentiate. Separation was comparable with and without aldehyde fixation.
