Human T cells maintain lifelong immunity to pathogens. However certain infections induce defective immune responses and involve CD8+ T cells dysfunction. Chronic infections in particular have been associated with CD57 expression by senescent CD8+ T cells. We sought to characterize the expression of CD57 by CD8+ T cells from healthy humans. This CD57 clone was selected based on personal communications from other labs.

PBMCs were obtained from healthy peripheral blood and lymphocytes were enriched following density centrifugation. Steady state analysis of T cells was then performed through multi-color flow cytometry. Cells were stained for surface markers to identify CD4/CD8+ T cells as well as CD57. Amongst CD8+ T cells, a clear population that was CD57+ was observed, even in healthy humans. Signal was retained following aldehyde fixation/permeablization.

Reasonable choice for multi-color panels involving CD57.
