Dendritic cells (DCs) mediate adaptive immune responses through the presentation of antigens to lymphocytes. DCs are heterogeneous and are found in different anatomical sites in both mice and humans. Expression of the integrin CD11c marks mature DCs and can be used to identify these populations by flow cytometry. We are interested in the development of mouse dendritic cells in vitro and in vivo and how metabolic signals may regulate this process. The mouse CD11c PerCP-Cy5.5 antibody was selected for multiplexing with a 10-color flow cytometry panel.

Results Summary

Spleens from C57BL/6 mice were harvested and homogenized to obtain a single cell suspension. Following red blood cell lysis, samples were stained to detect mature dendritic cell subsets. DCs were identified as CD11c+ MHCII+. A clear DC population was detectable and could be further analyzed for subsets specific markers. Staining and separation was maintained after aldehyde fixation/permeablization.
