Our goal was to establish conditions for highly reliable cytokine staining in a high throughput setting. I tested the same clone of anti-IL-10 conjugated to several of the brightest fluorophores currently available. This one provides a signal that rivals PE, with minimal non-specific binding and spreading of the negative population. Furthermore, this conjugate provided similar staining intensity even when the antibody concentration varied by over an order of magnitude (between 1:100 and 1:2000). This makes it highly resistant to minor changes in researcher handling.
