Nonspecific background amplification is a major problem during the setting up of a PCR reaction at normal room temperature.  IMMOLASE DNA polymerase from Bioline is a robust hot start and thermal stable polymerase which can be activated only by high-temperature incubation. So we can use this polymerase at room temperature without background amplification.

Protocol Overview

Roughly 100 ng of cDNA/50ng of DNA is used as a starting material. 2 microliter of the 5x immobuffer is added along with 0.5 microliters of the 10 micromolar dNTPs with gene-specific primers were added. Finally, 0.5microlitersr of the enzyme is added to the 20 microliter reaction set up. PCR is programmed with 10 minutes initial denaturation step, followed by 35 cycles of denaturation, annealing and amplification steps.

Tips

Initial prolonged denaturation step is required.

Results Summary

A very good amplification of the cDNA is obtained with no background amplification.
