After cell extraction and 4h in vitro re-stimulation, block your cells with Fc block, wash your sample 2x with washing buffer, stain for 30min RT with your extracellular staining, wash 2x and then proceed with intracellular staining protocol for Il-2 (1:200).The experiment was performed on TILs and spleenocytes of brain tumor bearing mice after immuno-therapy, to show change in cytokines expressions.
