Guava Cell Cycle Reagent for Flow Cytometry

I am testing 2 drug compounds for their ability to kill ovarian cancer cells through cell cycle arrest. We sought to measure the ability of these compounds to cause cell cycle arrest in SKOV3 cells either alone or in combination. I chose to try this reagent instead of traditional PI because we have a Guava machine in the lab that isn't being used.
Flow Cytometry, Cell Cycle Analysis
Ethanol-fixed SKOV3 cells
Following drug treatment, cells were pelleted in tubes and fixed in 100% ethanol overnight. Following a wash in PBS, pellets were resuspended in 200ul of Guava Cell Cycle Reagent and incubated for 30 minutes at RT (in dark). Data were acquired on the Guava instrument.
Be sure to completely wash out ethanol before staining!
We get excellent, reproducible results in untreated cells which gives us confidence that the observed cell cycle arrest data are accurate.

Inexpensive, fast and reproducible results
No drawbacks with the product, but the Guava instrument can be finnicky.
Excellent product to do rapid flow cytometry without having to pay core fees. Highly recommend.
