Usually the quantitative expression of the genes requires a standard reference gene to minimize the cellular, RNA quality and handling error. Generally, when handling serum sample RNA, we really don't have a standard deference RNA to normalize the data. ERCC ExFold RNA Spike-In Mix provides us with external RNA which can be added to the serum samples before isolating total RNA. This external RNA acts as the reference genes for analysis of gene expression using NGS.

Protocol Overview

During isolation of the RNA from the serum/plasma samples, a known concentration of the external RNA is added to the serum/plasma samples and total RNA is isolated from serum samples. Then we proceed for preparation of the RNA libraries for the samples.

Tips

You have to use this kit only you are working with serum/plasma/milk samples.

Results Summary

After preparation of the RNA libraries from the external RNA added total RNA, the libraries went on for RNA sequencing. Once the sequencing run is completed, the results must be mapped and counted using ERCC reference file.
