miRNA biomarker discovery
I used this kit with human serum samples. This kit can be used for samples of other species as well.
200 microliter of plasma is used as the starting material to which 5 volumes of lysis reagent is added along with a spike in control. Chloroform is added and vortexed followed a centrifuge of the samples at 13000 rpm. Then the upper aqueous phase is loaded on to the Minelute spine column. The column is washed with RWT and RPE buffer and finally with 80% ethanol. 12 microliter of nuclease free water is used for eluting the small RNA
Always use fresh serum/plasma samples for miRNA isolation as long storage reduce the yield.
A good quality of enriched miRNA are obtained in the final elution of column which can be quantified using a good cDNA synthesis kit.
