We are looking at various cytokines released in response to different in vivo stimulations. We used this assay to detect levels of IL-1alpha in various mouse tissues. We chose this product because most of our other ELISA kits and antibodies are by eBioscience, and we felt the pricing was reasonable.

Protocol Overview

Standard sandwich ELISA protocol - coat overnight, wash, block, add samples & incubate, wash, apply secondary, wash, apply Avidin HRP, wash, add substrate, add stop solution. Protocol can be done in 5hrs after overnight coating. Wash solution must be azide-free.

Tips

Fairly easy to use; works best when samples/standards are incubated overnight.

Results Summary

Generates 450 OD values, works well on multiple plate readers. Sensitivity ~5pg/ml - 500pg/ml.

Easy to use, everything you need (minus wash buffer) comes with kit. Produces consistent results.

Nothing, really. We've seen some variation in the standard curve, but that is likely due to technique.
