Our lab needed an adult human fibroblast cell line to compare with our human fetal fibroblast cell lines for ongoing apoptosis and fibrosis studies. Cell Applications had been recommended to us and been referred as very reliable. We have been using the cells now for a few months and are very satisfied with the results so far. The kit comes with the required medium and subculture reagents for establishing and maintaining the cells. Cells have been easy to culture and propagate and have acquired the estimated 12 population doublings as company states.

Protocol Overview

Followed Cell Application's recommended Sub-culturing HLF Protocol, and used the subculture reagents included in the kit of HBSS, Trypsin/EDTA & Trypsin Neutralizing Solution.

Tips

Important to rinse at least 3 times with HBSS which allows for easier lifting of cells from surface. Also, do not expose to trypsin longer than 4 minutes.

Results Summary

Cells attached, spread, and proliferated as expected in Growth Medium provided in the kit. Fibroblasts were cultured and propagated to at least 12 doublings and a little beyond without losing morphology characteristics.

Summary

The Good

Everything you need to culture and maintain cells is included in the kit. Had no problem culturing and maintaing cells.

The Bad

Would have liked to extend use of cells well beyond 12 doublings, but realize this is hard to acquire with adult cells. Noticed had to only used recommended company supplied media as cells flourished in this formula much better than others we've used with our established fetal fibroblasts.

The Bottom Line

These adult HLF's provided the necessary cell line we needed to compare with the fetal fibroblasts in our studies. Easy to culture and maintain and a reliable source for future cell lines if needed.
