We studied the mRNA expression of CD320/TCblR and megalin in mouse brain. We used WT and CD320/TCblR KO mouse. The studies were done to determine whether megain mRNA levels change in absence of TCblR.
Measuring mRNA extracted from mouse brain tissue

Tissues collected were homogenized in Trizol and mRNA extraction was done prior to synthesize cDNA. We used 5 micrograms of RNA for cDNA synthesis. Manufacturer's protocol was followed.
Make sure the RNA is of good quality
cDNA prepared was further used to run qRT-PCR. We found slight reduction of megalin mRNA in TCblR KO mouse.
