I battled with isolating quality DNA from a complex human biofluid. I tried this kit first. I found that it was constantly clogging despite cleaning up the sample before hand. Spinning resulted in a terrible yield of DNA and it was not quality DNA. I got a much better yield when I used a vacuum pump to pull the sample through the filter. I have used this kit before, but if you have even a slightly dirty sample you will find that this kit is not for you. Instead I would recommend the ZYMO DNA kit. It was able to handle a dirty sample, resulted in exceptional yield, and was pure.

Add your sample to a tube, add lysis buffer add buffer to bind DNA and spin, wash with 2 different wash buffers spinning between each wash, then elute DNA.
Use a vacuum pump instead of a centrifuge to pull the sample through the filter, also do not use this kit with any sample that is slightly 'dirty'
Overall the yield was unimpressive compared to other kits and the final DNA product was not pure.

On a pure very clean sample this works well. For anything somewhat complex or 'dirty' avoid at all costs
