We are studying our protein of interest interacting partners in glioma. To find out the interacting partners, we needed to execute the pull-down assays and perform immunoblot analysis. Manually developing blots is difficult in a long run, so we are using a luminescent image analyzer.

Western transferred nitro cellulose membrane
Before your blot is ready for probing with ECL solution, switch on the Las 500. It will take 5 min for it to get ready. Two plates will come along with the machine. Keep your western blot on one plate and the other plate is for the agarose gels to get pics. You need to keep your nitro cellulose membrane on the thin plate and add the ECL solution. Now you can take the images by manually by setting the program to take images at different time points.For taking images from agarose gels for DNA, we have to keep one red filter in the middle and then place your agarose gel on a thick aluminum plate to take the pictures manually.

We got very good results and find some novel interacting partners without protein of interest.
