We study a KO mouse model of cobalamin deficiency and have used M.O.M. kit to perform immunohistochemistry using a mouse monoclonal primary Ab. It worked great with almost no background.

Protocol Overview

Prior to blocking, we incubated the sections with 3% hydrogen peroxide. The rest of the protocol was followed exactly the way it is provided by the manufacturer. We incubated the sections with primary antibody for 30-60 minutes. A control was set with no primary Ab added to the reagent that was used for primary Ab dilution.

Tips

Make a fresh dilution of all the reagent whenever required.

Results Summary

We obtained immunostaining as desired with almost no background in mouse hippocampus. The results showed clear differences between WT and KO mouse staining indicating lower expression of the protein of interest in the KO mouse.
