Mature centrioles have assessor structures (called distal and subdistal appendages) attached near their distal ends. My work attempts to elucidate the function of the subdistal appendages of the centriole. We double checked the localization of Cep128 and centriolin to confirm that they localize to the subdistal appendages. We also needed these reliable antibodies to confirm depletion of the proteins with RNAi and CRISPR. The antibodies also allowed us to determine which conditions lead to the mislocalization of these proteins (Cep128, Centriolin, ODF2, ninein,…).
Immunofluorescence
Tissue Cultured human cells fixed with either Methanol or 4% PFA
1:400 dilution, 1 hour
3%BSA, 0.1% triton in PBS with sodium azide
Goat anti-Mouse Igg1 (Molecular Probes)

Flourescent Microscope, 100x objective
Antibody works reliably to stain the centriole's subdistal appendages. Works with both PFA and methanol fixation. Can stain both human and mouse cell subdistal appendages.
