We frequently carry out colorimetric assays to monitor the growth of our organisms. We use this instrument to measure absorbance readings of the assays.
Colorimetric Assay
Archaeal enrichment culture, water, Greiss reagent
A small amount of culture is diluted in water, followed by addition of the Greiss reagent. After 5 minutes, the color change is measured using the spectrophotometer.
The displayed absorbance reading may change from the first number shown. Wait for the display to blink three times before recording the reading.
Based on the measured absorbance of the assay, we are able to determine the concentration of metabolite produced in the culture and monitor its growth.
