Alexafluor 488 Phalloidin was used to stain actin in order to quantify actin structures such stress fibers, filopodia and lamellipodia in fixed cells. By treating cells with various compounds or by reducing levels of specific proteins by siRNA treatments, the affects on the actin cytoskeleton can be visualized using this product. As phalloidin is a chemical stain, that is highly specific to actin, it is superior to antibody staining and far simpler to use.
F-actin staining of fixed mammalian cells
Fixed mammalian cells. Examples: Human U2OS cells, HUVECs, Hacats and HeLa. Mouse NIH3T3's and primary fibroblasts
This product is used at a 1:200 dilution (in PBS). 30 minutes of incubation is sufficient for complete F-actin staining.
Can be used in combination with immunfluorecent staining procedures.
