We wanted to determine the number of CD4 positive cells in the spleens of our newly developed mouse strain by flow cytometry.

Mouse Spleen
Single cellswere resuspended in 0.2ug of CD4 antibody diluted in buffer. Cell mixture was incubated at room temperature for 1 hour.
1 ug Fc Block (purified 2.4G2 antibody directed against FcgII/III receptors) was used for 15 mins at 4 degrees celcius
PE Streptavidin diluted in incubation buffer was used for 30 mins at room temperature

We were able to consistently detect cells expressing CD4 in all of our cell suspensions.
