We wanted to determine the concentration of IL-12 secreted by bone marrow derived dendritic cells co cultured with a potential cytokine inhibitor.

I followed the standard sandwich ELISA protocol provided by the manufacturer. The kit contained all of the components needed to perform the ELISA including the capture antibody, detection antibody, Streptavidin-HRP and Substrate solution. All components were sequentially added to a 96 well Nunc plate, with the sample between the capture and detection antibodies. All wells were wash 3x between solutions. A microplate reader set to 450 nm was used to measure the optical density.
Label and prepare all reagents before beginning.
We were able to determine the concentration of IL-12 in all of our samples.
