We routinely purify different cell types for in vitro killing assays where effector cells are tested against HIV infected target cells. We are expressing different TCR vectors in human CD8 T cells to reprogram their recognition of MHC I - peptide on infected cells. These vectors have a mouse CD8 reporter to distinguish the expressed TCR from the endogenous and we use a two step labeling, first with mouse CD8 antibody, then with biotin beads, to purify the transduced cells
