Flow cytometry is a common technique in our lab which studies CD4+ T cells in inflammatory processes both in vitro and in vivo. To minimize the use of precious sample when performing compensation, we often use compensation beads.

Experimental Design and Results Summary

Application

Flow cytometry

Starting Material

UltraComp eBeads and flow cytometry antibodies

Protocol Overview

Label tubes for each antibody that is going to be used for flow cytometry, add ~200-300ul flow buffer, add one drop of beads to each tube, add flow antibody at dilution used for staining experimental samples.

Tips

Be sure to have a tube of unstained beads

Results Summary

Compensation is achieved without the use of experimental cells/sample.

Summary

The Good

No experimental cells/sample is used when performing compensation

The Bad

Have to adjust FSC and SSC and gates to see bead populations during acquisition

The Bottom Line

Great product for anyone doing flow cytometry with experiments with limited cells/tissue.
