Normalization of western blot loading is critical when assessing protein levels in a given sample. LI-COR offers a water-soluble fluorescent stain to quantify total loaded protein for western blotting. Following transfer, blots are stained with the kit and total protein can be imaged on a LI-COR western blotting machine. The dye can then be washed out and blot can be blocked and put in primary. Total protein for each sample on the blot can be quantitated using LI-COR imaging software.

Protocol Overview

Rinse nitrocellulose membrane after transfer with water, incubate membrane in 5mL Total Protein Stain for 5min with gentle shaking, drain stain and wash membrane twice in wash solution, image membrane in 700 channel on imaging system, block membrane and blot as usual

Tips

Be sure to thoroughly wash membranes before moving on to blocking step

Results Summary

Total protein can be visualized in much the same manner as performing a Ponceau stain
