Our lab routinely performs the EAE model in C57Bl/6J mice which requires the injection of an emulsion of myelin oligodendrocyte glycoprotein (MOG) peptide and Complete Freunds Adjuvant (CFA). In order to make the CFA, heat-inactivated M tuberculosis in ground into incomplete Freunds adjuvant.
In vivo injections in mice (EAE model)
Incomplete Freunds adjuvant and heat inactivated M tuberculosis
Grind heat inactivated M tuberculosis with incomplete Freunds adjuvant for ~10 minutes using a mortar and pestle
Use a mortar and pestle to grind M Tb into the incomplete adjuvant
Mice injected with CFA+MOG develop EAE
