We develop siRNA therapeutic for cancer. In this scenario, we needed to screen the level of PLK1 protein in colon cancer cell lines and determine whether our siRNA has good knockdown. Cell Signaling 4513 was compared to PLK1 antibodies from other manufacturers and performed better.
Western Blot
Human colon cancer cell line lysate, 20ug loaded per lane.
1:500 in Licor blocking buffer diluted 1:1 with TBS, contained 0.1 % tween 20, overnight at 4 degrees.
Licor blocking buffer
Licor IRDye 800CW, 1:10,000 in Licor blocking buffer diluted 1:1 with TBS, contained 0.1 % tween 20 and 0.05% SDS, 2 hours at room temperature.

PLK1 protein level was detected across 8 colon cancer cell lines, in which their expression level agreed with existing published data (A). Knockdown was achieved with our siRNA and properly detected (B). Specificity was good as we did not observed any additional band on the membrane.
