Really happy with the results, actually managed to use AmCyan Channel for a positive staining! Given, its CD45, but the staining looked great and opened up another channel, so I am happy with it.

Results Summary

Staining looks great, clear CD45 populations, including PMN (larger SSC). Previously, I had never gotten AmCyan to work in my panels, and avoided it whenever possible, and instead had CD45 in Alexa700 because I had less issues with compensation. However this CD45_BV510 was incorporated without any serious issues, and seems brighter than Alexa700. I did have more complications in my staining panel (more compensation issues and just weird flow laser artifacts), so I still don't love this color / channel. However, its great for CD45 and I'm happy I can use it, allowing for another channel in my mixes. I don't know if I would trust it for any other antigens but CD45 is prevalent and not cross compared with other colors in the way I do my gating, so I'm overall very pleased with the results.

Summary

The Good

CD45 BV510 gives clear populations

The Bad

BV510 and AmCyan have issues with spillover, requires increased compensation and careful panel design

The Bottom Line

Acceptable result and works in my panel. Pleased with the results.
