I aimed to assess the effects on regulatory T cells (Treg) in my germline knockout mouse. Measure the frequency of Treg in total CD4 T cells is the 1st step to examine the consequence of loss my study gene. Biolegend's True-Nuclear Transcription Factor buffer has been optimized for surface and intracellular staining process.

Mouse spleen cells

Protocol Overview

1.Surface staining: CD4-PE (1:500) + TCR Beta-ACP/Cy7 (1:300) at 4C for 25 mins. Then wash 1x with 1ml FACS buffer.2.~ 300ul Fix/Perm buffer at RT for 20 mins. 3. 1ml FACS buffer wash 1x to remove Fix/Perm buffer.4.~300ul Biolegend Perm buffer at RT for 15mins. Then wash with 1ml FACS buffer to remove Perm buffer5. Resuspend in ~90ul Perm buffer, staining Foxp3-488 (1/300-) at RT for 25 mins.6. Wash 2x with 1ml FACS buffer and then process to FACS analysis. 7. Resuspend in ~400ul FACS for FACS analysis.

Tips

Freshly prepared Fix and or Perm buffer before experiments.

Results Summary

I can detect very clear separated Foxp3- and Foxp3+ T cell populations. The frequency is similar to Foxp3 transgenic mouse results.
