Our laboratory investigates different ways to improve immune responses against HIV.. We developed different mouse models to study the efficacy of therapeutic candidates against HIV. Some of these mice express human cell surface receptors utilized by HIV to enter and infect the cells. We use an infectious HIV virus expressing nano luciferase to infect these mice and study different therapeutics that might increase or suppress infection. We use nano luciferase because it is much brighter than Renilla luciferase and allows us to more precisely detect differences in infectious between different treatment groups. We use this kit to detect nano luciferase at the endpoint of our experiments.

Mouse cells of tissue

Protocol Overview

Isolate the cells from mouse tissue of interest. Count and lyse the cells (using passive lysis buffer by Promega). Prepare the nano luciferase substrate. Incubate cell lysate with substrate. Read luciferase values on a luciferase machine.

Tips

In our experience nano luciferase reporter is not optimal for in vitro use due to high background.

Results Summary

High nano luciferase values correspond to high infection values.
