We are interested in enhancing the killing capacity of HIV-specific CD8 T cells by reengineering TCRs expressed by these cells. We evaluate TCRs cloned from CTLs of infected patients for their ability to elicit CTL response by first transducing a Jurkat/MA cell line which has a NFAT driven luciferase reporter. We can directly measure TCR activation by using this kit to detect luciferase activity.

Lyse the cells using the promega lysis buffer (5x). Prepare the substrate by combining a vial of assay buffer (10ml) with a vial of lyophilized substrate. Mix 100ul of cell lysate with 100ul of luciferase substrate. Read samples on luciferase machine,
The hydrated substrate can be stored in -20 protected from light for several months without the loss of activity.
