Our laboratory investigates different ways to improve immune responses against HIV, including enhancement of the capacity of effector cells, such as NK cells, to kill HIV-infected cell. In our experiments we use infectious HIV virus encoding for Renilla luciferase reporter. We use this assay to measure levels of luciferase activity as an indication of HIV infection.
Luminescence measurment
Cells infected with HIV-LucR virus
Count cells, Lyse using the promega lysis buffer. Prepare luciferase substrate by mixing the assay buffer and substrate (100x). mix 20ul of cell lysate with 100ul of substrate. incubate for 30s. Read on a luciferase machine.
Follow protocol
high luciferase values correlate with positive control for HIV infection. See a decrease of infection when cells are treated with known inhibitors of infection.
