We are interested in enhancing the killing capacity of HIV-specific CD8 T cells by activating them via TCR specific MHC-peptide ligands, and co-stimulatory molecules. We use purified CD8 T cells from naive donors. We do different FACS experiments with these CD8+ T cells, including staining for activation markers and cytokines after co-culturing the CTLs with cells presenting peptides specific for the TCRs expressed by the CTLs. In all of these experiments we stain for hCD8 so that we can gate on the CTLs specifically. We use this antibody conjugated to different fluorophores to use in different panels.
