We are interested in enhancing the killing capacity of HIV-specific CD8 T cells by reengineering TCRs expressed by these cells. We use purified CD8 T cells from healthy donors to transduce with the different lentiviral constructs, and use the LentiBlast kit to improve transduction efficiency.

Protocol Overview

Cells are transduced with the desired construct with 0.5 microliters of solution A and 0.5 microliters of solution B per ml. Cells are plated in a 48 well, and spun down for one hour.

Tips

Make sure you titrate the volume needed for each solution.

Results Summary

Great transductions: 30-40% for primary cells, which is difficult to get. Sometimes polybrene can give the same results, but it's more toxic to the cells.
