Our lab uses HIV molecular clones that express Nano luciferase reporter. This reporter is very stable and allows quatification of low HIV infection, because it provides a very sensitive reading. We infect cells in vitro, or perform in vivo experiments where we infect our mice with these viruses. As a read out, we test cell lystaes for their luciferase activity by using this kit.

Cells are lysed in 1x lysis buffer, vortexed well, stored in -80 degrees for at least one hour, thawed, then read in presence of substrate.
