Our lab has developed an in vivo humanized mouse model consisting of injecting human PBMC into NSG (immunocompromised) mice to study HIV pathogenesis and test therapeutic agents. Studies investigating different latency reversing agents and treatments involve a detailed analysis of different cell populations in the spleen including memory and effector CD8 and CD4 T cells. Antibodies distinguishing these populations are needed for this purpose.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Human PBMC gated on CD8 T cells

Primary Incubation

1:100 for 30 min at 4 degrees Celcius

Blocking Agent

None

Secondary Incubation

None

Tertiary Incubation

Detection

Flow cytometry

Results Summary

Antibody works. It is hard to gate on positive and negative populations given that the transition is not very distinct, but with good controls it is possible to separate these populations correctly.
